Goal: We suspect the antibiotic “X” resistance gene is located on large plasmids in P. aeruginosa. We need to isolate bacterial plasmids for further analysis.

Cell Growth:

1) Select one of the P. aeruginosa colonies that grew on the antibiotic “X” (+) medium, and use this to inoculate 5ml of TSB medium in at 18 X 150mm test tube. Grow the culture overnight (16-18 hours) at 30◦C using a tube roller set at 100-200 rpm.
